Glycine and sleep: what the evidence actually shows

Glycine is the smallest amino acid in the body, and three grams of it before bed is one of the more interesting ideas in sleep nutrition. Small trials report falling asleep faster, reaching deep sleep sooner, and feeling clearer the next day. The evidence is real. It is also young, modest in size, and mostly the work of a single research group, which is exactly the sort of thing a supplement company should tell you before selling it to you.

What is glycine?

The simplest amino acid there is, and one of the most abundant in you right now. Around a third of collagen, the protein that holds skin, tendon and bone together, is glycine. Your body makes it, food supplies more, and it tastes gently sweet, which is unusual company for something with a role in the nervous system, where it acts as a calming, inhibitory neurotransmitter in the brainstem and spinal cord.

None of that, on its own, explains why swallowing a few grams of it at 10pm should change your night. The proposed explanation is more specific, and it is the reason we take glycine seriously.

How could an amino acid help you fall asleep?

Through temperature. Falling asleep is a thermal event as much as a mental one: sleep onset normally follows a drop in core body temperature, achieved by sending warm blood to the skin of the hands and feet. In a well-known Nature paper, the warming of hands and feet was among the strongest predictors of how quickly people fell asleep [1]. Glycine appears to act directly on this mechanism. Work in Neuropsychopharmacology traced its effect to NMDA receptors in the suprachiasmatic nucleus, the brain's central clock, where it triggers blood flow to the extremities and a measurable fall in core temperature [2]. In other words, glycine seems to press the same physiological switch a warm bath or bed socks press, chemically and from the inside.

A mechanism is a hypothesis, not a result. So what happened when people actually took it?

What do the human trials show?

Three small Japanese studies form the core of the evidence, all using 3 grams before bed.

The first, in 2006, gave glycine or placebo to volunteers with ongoing sleep complaints and found improved ratings of sleep quality and morning clarity, measured by questionnaire [3]. The second, in 2007, is the one that matters most, because it put participants in a sleep laboratory. Glycine improved subjective sleep quality and sleep efficiency, and shortened the measured time both to sleep onset and to slow-wave sleep, without distorting the overall architecture of the night [4]. That last detail deserves a pause: sedating drugs typically buy sleep by changing its structure, and glycine did not appear to.

The third took healthy people and did something crueller: cut their time in bed by a quarter for three nights running. Those on glycine reported less daytime fatigue and performed better on vigilance testing than those on placebo [5].

The direction is consistent and the mechanism is plausible, and one of the results came from a sleep laboratory. The numbers are small and the durations short. There is one more thing you should know.

Who ran these trials, and does it matter?

The core studies share authors affiliated with Ajinomoto, a major amino acid manufacturer with a commercial interest in glycine. That is worth saying plainly. Industry involvement does not make findings false, and much nutrition research would not exist without it; the mechanism work is careful and the laboratory study measured objective outcomes that are hard to wish into existence. But it does mean the evidence base and the commercial interest grew up in the same house, and the honest response to that is to weight independent work more heavily as it arrives.

A 2024 systematic review took stock of glycine research across the body, covering around fifty human studies, and its conclusion carried the same double edge. The nervous system, and sleep in particular, showed glycine's most consistent positive signal of any domain, and the underlying sleep trials were judged small and at high risk of bias [6]. Both halves of that sentence are true at once, and we would rather quote the pair than the flattering half.

Independent signals are beginning to appear at the edges. A 2024 crossover trial in physically active men with sleep complaints, from a separate research group, gave a glycine-rich collagen peptide drink before bed and recorded fewer night awakenings on polysomnography along with better next-morning cognition [7]. It used collagen peptides instead of pure glycine, so it is adjacent evidence and stops short of replication, but the direction rhymes.

What dose, and when?

The studied dose is 3 grams, taken within the hour before bed. That is a genuinely awkward amount for the supplement industry, because it does not fit in a capsule or two, which is why glycine so often appears in sleep products at token doses of a few hundred milligrams. A sprinkle of glycine lets a label mention it. It is not what the trials tested.

As a powder the dose is easy, and glycine's mild sweetness makes it one of the few actives that improves a drink instead of ruining it.

On safety, glycine is a normal component of the diet and 3 grams sits well within amounts the body routinely handles; the trials above reported no meaningful side effects at this dose. The sensible caveats are the standard ones: if you are pregnant, breastfeeding, or take prescription medicines, have a word with a pharmacist first, and no supplement is a treatment for insomnia disorder, for which the front door remains your GP and CBT-I.

So where does glycine honestly land?

Somewhere refreshingly specific. If your problem is switching off at bedtime, glycine at the full 3 gram dose is a reasonable, low-risk thing to try, backed by a coherent mechanism and small consistent trials, and you should expect a modest effect at most. Common questions

How much glycine should I take for sleep?

The dose used across the human trials is 3 grams, taken within an hour of bed [3][4][5]. Products containing a few hundred milligrams are quoting the research while not matching it.

How quickly would I notice anything?

The trials measured effects from the first nights of use, so this is not an ingredient that needs weeks to build up. If a fortnight of proper dosing changes nothing, that is a fair trial and an honest answer.

Can I just get glycine from food?

You already do, especially from collagen-rich foods, spread across the day. The trials tested a single concentrated dose shortly before bed, timed against the body's temperature drop, which ordinary meals do not replicate.

Is it safe to take every night?

At 3 grams, glycine has been well tolerated in the trials to date, and it is a normal part of the diet rather than a foreign compound [3][4][5]. Speak to a pharmacist if you are pregnant or on regular medication, and see your GP if poor sleep is persistent, since long-term insomnia deserves proper assessment, not indefinite self-treatment.
